Upstream Principal Scientist, Vector Biology

Legend Biotech is seeking a Principal Scientist for their Vector Biology Upstream team in Philadelphia, PA. This role focuses on early process development for lentiviral vector (LVV) platforms to support in-vivo CAR-T programs. The scientist will lead the definition, optimization, and scaling of upstream LVV processes from construct design to bioreactor manufacturing.

• Establish and optimize early-stage seed train strategies from shake flask through bench-scale bioreactor
• Apply molecular biology expertise to evaluate LVV constructs for manufacturability and potency
• Design and optimize LVV production workflows, including transfection and stable producer cell line platforms
• Conduct media optimization, feeding strategy evaluation, and bioreactor process control development
• Partner with Downstream colleagues to integrate upstream design decisions for end-to-end process coherence
• Design and execute early process development studies using Design of Experiments (DoE)
